Abstract
The purpose of this study was to isolate secondary metabolites, determine the toxicity and antimicrobial activity of extracts and compounds from the bark of M. umbellata. The process of extraction, isolation and purification of secondary metabolites from the stem bark of M. umbellatawere carried out by means of maceration, chromatography techniques, and crystallization and recrystallization. The toxicity test using the BSLT method used Artemia salina, then the antibacterial and antifungal tests used the agar diffusion method (Kirby-Bauer diffusion). Based on the spectra data from FTIR, UV-Vis, NMR measurements (1D and 2D NMR), the compound isolate was identified as 3β-acetyl-olean-12-en-28-oic acid. The results of the toxicity test showed that the 3β-acetyl-olean-12-en-28-oat acid compound, methanol extract, hexane, chloroform, and ethyl acetate, from the bark of M. umbellatawere toxic to A. salina, but methanol extract was the most toxic against A. salina with an LC50value of 169.863 ppm. Ethyl acetate extract and 3β-acetyl-olean-12-en-28-oat compound, at a concentration of 1000 ppm were able to inhibit the growth of S. aureus bacteria with a resistance zone diameter of ˃ 14 mm. Methanol extract has inhibitory power against the growth of C. albicans fungi with an inhibition zone diameter of 10, 48 mm. Meanwhile, chlorophore extract and hexane showed weak inhibitory power against tested bacteria and fungi with an inhibition zone <10.00 mm
