Uji Toksisitas Akut Larutan Rimpang Temu Kunci (Boesenbergia pandurata) Dan Pengaruhnya Terhadap Perubahan Histopatologik Insang Ikan Mas (Cyprinus Carpio L)
Abstract
The aimed of this research was to determine the level of acute toxicity of the finger root (Boesenbergia pandurata) solution, to analysed histopathological changes of gill , swimming pattern and respiratory frequent on common carp (Cyprinus carpio L) was exposed the finger root solution. The research was conducted in January - June 2020 at the Laboratory of Aquatic Toxicology, Faculty of Fisheries and Marine Sciences, Mulawarman University. The Acute toxicity test procedures described in USEPA (1992) consists of Preliminary test has to required upper and lower limit test concentration were 1000 mg.l-1 and 100 mg.l-1 and Definitive test to determine 96 h-LC50 value has been used five concentrations namely 100 mg.l-1, 125.89 mg.l-1, 199.50 mg.l-1, 316 mg.l-1, 500.88 mg.l-1 and control. All treatments with 3 replications. Preparation of Histopathological made by paraffin method, fixative with bouin solution and stained with Hematoxylin –Eosin, 96h-LC50 value analysed with Probability Unit (Probit), histopathological changes of gill analyses descriptively, swimming pattern and respiratory frequent with visual analyses. The result of this research showed that the 96h-LC50 of finger root solution was 274.41 mg.l-1is mean finger root solution could be affected to the 50% mortality animal test with concentration 274.41 mg.l-1 and criterion was moderately toxic. The histopathological changes of gill caused of exposed in solution were oedema, hyperplasia, haemorrhage, necrosis and fusion of lamella secondary. Swimming pattern was change and the frequency of respiration was increased after exposed finger root solution during 96 hours
